A couple of dynasty years back, I started really using this option to strengthen my offense, and it is really working out well. This is what I do.
I run a lot out of the I-Normal and Ace-Normal, and prefer to run to the right. So, when in those formations, I put my WR and TE who are best at run blocking, and/or are bigger physically (usually one in the same) on that side. I get far better blocking performance, especially on the corners.
In passing formations, I put my best WR on the far right side, away from the best CB for the defense. On the wide left, I use a big receiver with good hands. The slots go to my speed guys, since they are the ones with great potential to be covered by a LB or S. You can also sub in better pass blocking linemen for shotgun / 4 or 5 wide sets, and conversely use the better run blockers for the I, Ace, or whatever else you run out of.
If at anytime you want another guy in, for a certain play, it is easy enough to do, but the game is much more fluid with formation subs, and it is easier not to forget, or get lazy and decide to stick with the guy out there.
Furthermore, if you have depth, you can put equal quality WRs in different formations so that they stay fresher, longer, and perhaps screw the defense up.
It also works really well for defense.
